       Charcoal-Stripped, Fetal Bovine Serum (CS-FBS) is used to elucidate the effects of hormones in a variety of in vitro systems. Hormones act as chemical messengers to different bodily systems and/or organs stimulating general and specific effects on the body and retarding others. Such processes such as growth, reproduction, sexual attributes, behavior and control of metabolic processes are dependent on hormones. The total number of hormones is still undetermined but each plays unique roles and has diverse functions. After a hormone is released from its so-called master gland, cell, or tissue into the capillaries or lymphatics, it may travel a circuitous route via the bloodstream to exert influence on not only other cells and tissues but also target organs far removed from its site of origin. Steroid hormones are derived from the blood lipid, cholesterol. The hormones such as testosterone, estradiol and cortisol differ only slightly in their chemical structures, yet have widely different effects on the body. The steroid hormones are secreted by the adrenal cortex, ovaries, placenta and testes. Steroid hormones are lipids themselves and are thus termed, lipophilic. Due to the fact that these hormones are not H2O-soluble, they do not dissolve in plasma but rather travel in the bloodstream attached to protein carriers. When the hormones arrive at their target cells, they dissociate from their carriers and pass through the plasma membrane of the cell. Other steroid hormones then bind to very specific receptor proteins in the cytoplasm, while still others, travel directly into the nucleus before encountering their receptor proteins. Intact FBS with its often unidentified growth-promoting components (e.g. hormones, growth factors, steroid levels, etc.) may contain higher levels of hormones, growth factors and immunoglobulins as opposed to other kinds of Sera and thus under certain circumstances and in specific applications, may retard the in vitro development of some cells in culture or competitively regulate the growth of other responsive cells in vitro.

 

Studies utilizing CS-FBS include multi-faceted aspects of steroid-receptor binding, steroid regulation of cellular receptors, hormone secretion of various tissues and the function of thyroid hormones. The production procedure includes the use of charcoal and dextran to remove hormones and growth factors from the FBS. By reducing the levels of steroid hormones and growth factors, this type of serum has been shown to be effective in receptor studies or estrogen-related research and as such is used whenever reduced levels of hormones, growth factors and steroid levels are desired.

 

CS-FBS is especially utilized as a supplement for growth medium in unique and/or specific applications.  Two major advantages come to the fore. Handling & Stability:  

Storage conditions: -20°C

Serum should never be taken from a frozen or refrigerated state and placed in a water bath to expedite the thawing/warming procedure. Such handling compromises the serum quality and may cause the bottle to break. After the serum has reached room temperature, it may be then pre-warmed slowly to 37°C while mixing the contents thoroughly before adding to the medium of choice.
