【Scope of application】

【Instructions】
After the serum is taken out from the freezing area (do not directly thaw the serum from -20°C to 37°C, as the temperature changes too much, it is easy to cause protein agglutination and precipitation), put it at 2°C to 8°C to thaw it. During the thawing process, shake gently and evenly intermittently, and then divide the package or use it directly after it is completely dissolved. Completely thawed serum is taken out from 4°C, and the basal medium is added in the required proportion (5%-20%) to form a complete medium. It is recommended that it be stored in a refrigerator at 4°C and used up within 1-2 weeks.

【Storage conditions and shelf life】

【Precautions】
The substances precipitated in serum mainly include: fibrin (flocculent precipitation), lipoprotein, cold agglutinin, vitronectin, fetuin, calcium phosphate (small black spot precipitation under the microscope), cholesterol, etc.

What are the causes of serum precipitation:

1. If the temperature changes too much, the serum should be thawed directly from -20°C to 37°C, which may easily lead to precipitation due to severe temperature changes.

2. If the serum is kept at room temperature or in a 4°C refrigerator for too long, the unstable components in the serum will be destroyed, affecting the cell growth effect; resulting in poor serum quality and precipitation.

3. When the serum is frozen and stored in aliquots, be sure to shake it evenly without generating air bubbles to avoid precipitation due to changes in the concentration of serum components.

4. Serum is prone to precipitation after inactivation. If you need to use inactivated serum, please follow the principle of 56°C, 30 minutes, and slight shaking. If the inactivation temperature is too high, the time is too long, and the shaking is uneven, precipitation may occur.

5. If there is serum precipitation, centrifuge after aliquoting. It is recommended to centrifuge at 400g for 5 minutes and use the supernatant.
